Benchmarking Takyon® Etheris
Takyon® Etheris removes background signals
Fig. 1A. Amplification curves with (line) or without (dotted line) E.coli gDNA (500 pg) in a qPCR reaction using either Takyon® Etheris Probe (orange) or a competitor (blue) kit. Each assay was performed in technical triplicates. All reactions were run according to the respective manufacturer’s protocols.
Fig. 1B. Mean Cq values obtained with Takyon® Etheris Probe qPCR Master Mix (orange) and a competitor kit (blue) in a 23S-targeted qPCR assay. Each reaction was performed in triplicate under identical assay conditions, either with 500 pg of E. coli gDNA or as a no template control (NTC). All reactions were run according to the respective manufacturer’s protocols.
The results indicate that Takyon® Etheris delivers higher sensitivity and stronger amplification signals, with no detectable signal from residual bacterial gDNA. By contrast, the competitor kit produced a Cq signal in the no template control (NTC), demonstrating contamination from residual host DNA.
Takyon® Etheris outperforms specialized competitor’s reagents
Compared to the competitor kit, Takyon® Etheris demonstrates improved sensitivity and stronger fluorescence signal intensity across tested targets, proving its value in multiplex conditions.
Fig. 2A. Amplification curves obtained with Takyon® Etheris Probe qPCR Master Mix (orange) and a competitor kit (blue) in a triplex assay targeting 18S, B2MG, and ACTB genes. Each reaction was performed in triplicate using 5 ng of DNA template according to the respective manufacturer’s protocols.
Sensitivity comparison of Takyon® Etheris vs competitor qPCR kits
Fig. 2B and Fig. 2C : Amplification curves obtained with Takyon® Etheris Probe qPCR Master Mix (orange) and a competitor kit (blue) for the amplification of the human ACTB (Fig. 2B) and 18S (Fig. 2C) genes. Five serial dilutions of genomic DNA were tested: 50 ng, 5 ng, 0.5 ng, 50 pg, and 5 pg. Each point represents the mean Cq value of triplicate reactions performed under manufacturer’s protocol instructions.
The data demonstrate that Takyon® Etheris delivers superior sensitivity and consistency across both ACTB and 18S targets, maintaining efficient amplification and reliable detection down to 5 pg of human gDNA. In contrast, the competitor kit exhibits delayed amplification and higher mean Cq values, indicating lower sensitivity and reduced amplification efficiency across gene targets.
Takyon® Etheris maintains the performance of the Takyon® range
Takyon® Etheris and Takyon® Ultra exhibit similar sensitivity and amplification performance across all three targets at 50 pg of human gDNA. Mean Cq values are closely aligned between the two formulations, confirming their equivalent efficiency in detecting low-copy templates.
Fig. 3. Amplification curves obtained with Takyon® Etheris Probe qPCR Master Mix (orange) and Takyon® Ultra Probe qPCR Master Mix (blue) for amplification of three human targets: 18S, ACTB, and B2MG. Each reaction was performed in triplicate using 50 pg of human genomic DNA as template.
